I n d i c a t i o n
The product consists of microplate coated with genetically engineered ASFV recombinant antigen and HRP-labeled
antibody and other reagents. It uses the principle of indirect enzyme-linked immunosorbent assay to detect ASFV
antibodies in pig serum or plasma. Used for auxiliary diagnosis of African swine fever.
P r e p a r a t i o n
1. The test should be used with fresh swine serum or plasma, and the dosage is 5μL.
2. It is forbidden to use samples contaminated with bacteria, lipemia, hemolysis or jaundice.
3. The sample should not be stored at room temperature for more than 8 hours; if it is stored for more than 8 hours, it
must be stored at 2-8°C; if it is stored for more than 1 week, it must be stored at -20°C.
T e s t m e t h o d
1. The kit is placed at room temperature for 30 minutes. The 20-fold concentrated lotion is diluted 20-fold with
deionized or distilled water.
2. Add 100μL of sample diluent and 5μL of sample to each well of the test wells, mix gently; one well of blank
reference without liquid; two wells of positive and negative reference, add positive reference serum and negative
reference serum respectively 100μL/well. After attaching the sealing film, place the box in a 37°C water bath or a
constant temperature incubator and incubate for 30 minutes.
3. Discard the sealing film and wash the plate with a plate washing machine or hand, so that each wells is filled with
the washing liquid without overflowing (about 300 μL), the soaking time is 10 seconds, the plate is washed 5 times,
and finally in absorbent paper dry up.
4. Add 100μL of enzyme working reference solution to each well (except the blank wells), After attaching the sealing
film, place the box in a 37°C water bath or a constant temperature incubator and incubate for 30 minutes.
5. As in the above procedure 3, wash the plate and dry.
6. Add 50μL of substrate solution and coloring solution to each well in sequence, gently shake and mix well. After
attaching the sealing film, place the box in a 37°C water bath or a constant temperature incubator, and incubate for 10
minutes in the dark.
7. Discard the sealing film, add 50μL of stop solution to each well, gently shake to mix, measure the OD value with a
microplate reader at a wavelength of 450nm (adjust to zero with blank wells), and print the results.
Keep out of reach of children.
R e s u l t s
1. Sample OD value ≥ Cut Off value, judged as positive, sample OD value < Cut Off value, judged as negative.
2. Cut Off=3.1×Average OD value of negative reference (when the average OD value of negative is <0.05, it is
calculated as 0.05; when the average OD value of negative is ≥0.05, it is calculated as the actual value).
3. Under normal circumstances, the negative <0.10, the positive ≥ 1.0.
C a u t i o n s
1. Wear gloves and work clothes during the experimental operation, strictly improve and implement the disinfection
and isolation system, and all experimental wastes should be treated as infectious materials.
2. The stop solution is corrosive and should avoid contact with skin and clothing. If it is inadvertently contacted, rinse
immediately with plenty of tap water.
3. When the enzyme plate is taken out from the cold storage, it should be restored to room temperature before opening
the bag. Unused enzyme plates should be stored in a sealed bag with a desiccant.
4. The concentrated washing liquid is easy to crystallize at low temperature, and it needs to be restored to room
temperature to completely dissolve.
5. Each well needs to be filled with liquid during washing to prevent the free enzyme from being washed.
6. The samples used for testing should be kept fresh.
7. The test results must be based on the reading of the microplate reader.
8. Reagent components of different batches should not be mixed.
Store at 2~8ºC. Shelf life: 12 months.
