Usage and the Determination of Results:
(1) The antigen are diluted with the 0.5% phenol normal saline to 1:20. The serum
sample should be diluted to 1:12.5, 1:25, 1:50, 1:100 and 1:200 with the same diluent,
prepare the positive serum and negative serum reference standard.
(2) Separately take 0.5ml of the antigen and serum in a tube, stand for 24 hours at
37ºC, observe the result of the agglutination test.
(3) Determination of results:
If the serums of cattle, horses, deer or camels show ++ in 1:100 dilution ratio, the
result is positive. If the serums of cattle, horses, deer or camels show ++ in 1:50
dilution ratio, the results is suspicious.
If the serums of pigs, sheep, dogs show ++ in 1:50 dilution ratio, the result is positive.
If the serums of pigs, sheep, dogs show ++ in 1:25 dilution ratio, the result is
suspicious .
For the animals, the test results is suspicious, should test again after 3-4 weeks. If the
test results is suspicious again, the conclude of the test is positive.
Criterion for agglutination reaction:
++++ The thallus agglutinate and sedimentate absolutely, the liquid is clear 100%.
+++ The thallus almost agglutinate and sedimentate absolutely, the liquid is clear
75%.
++ There have obviously agglutination and sedimentation, the liquid is clear
50%.
+ There have clearly visible agglutination and sedimentation, the liquid is clear
25%
- There have no agglutination and sedimentation, the liquid is turbid uniformly.
