Usage Range:
The detection card can be use for test the avian influenza virus antibody in avian
serum. The results can be use for immune effectiveness evaluation or auxiliary
diagnosis.
Requirements for the samples:
Serum: Take 2-3ml blood and place in a clean and dry test tube by routine method.
Stand for 1 hour, after the blood clotted. Put the clotted blood sample in centrifuge at
4000r/min for 10 minutes, get the serum ( can also stand the blood sample for 2 hours,
wait the serum separate from the clotted blood). The serum must be clear, without
hemolysis, without contamination. The serum can be kept at 2-8ºC for short-term
preservation, and should be kept at -20ºC for long-term preservation.
Whole blood: the whole blood with anticoagulation can be use for test sample. (The
whole blood sample can not be freeze.)
Test procedure:
1. Open the aluminium foil bag, take the detection card out, and put on a flat and
clean countertop.
2. Take the blood sample by pipette, drop 1 drop (about 25μl) onto the injection port .
(observe if there are liquid flow out from the well.)
3. After 10 minutes, drop 2 drops of buffer solution (about 60μl) onto the injection
port.
4. Stand for 10-15 minutes at room temperature. The test result is invalid if over 30
minutes.
Attention: If add over dose sample or buffer solution, the result will be mistaken.
Determination of the result:1. Negative reaction: If in the observation port, the purple red line appear at the test
line area (T) and control line area (C) simultaneously. It means the antibody level is
lower than 1:16 HI titer.
2. Positive reaction: If in the observation port, the purple red line only appear at the
control line area (C). It means the antibody level equal to or higher than 1:32 HI titer.
3. Invalid: In the observation port, there is not purple red line appear at the control
line area (C).
The threshold of the detection card is 1:32. Dilute the serum sample and find the
maximum dilution ratio, at which point the test results show positive reaction.
According the dilution ratio calculate the antibody titer. For example, dilute a serum
sample by normal saline to 1:2, 1:4 or 1:8, then test the three serum sample. If the test
results of 1:2 and 1:4 dilution ratio serum sample show positive reaction, and the test
result of 1:8 dilution ratio serum sample show negative reaction. Then the maximum
dilution ratio is 1:4, The final antibody titer of the sample is 2(5 1:32)multiply with 22
(1:4), equivalent to 27(1:128).
Attention:
1. Please read the instruction carefully before the test. All the reagents in the product
only can be use in the test.
2. The expired products or the aluminium bag broken products can not be used.
3. Take the detection card out from the refrigerator and stand for to room temperature,
open the aluminium bag, use as soon as possible, avoid invalid after dampness.
4. The deionized water, tap water and normal saline can not be use as negative
control.
5. The serum sample should be fresh and clear. Contaminate turbid sample, with
hemolysis sample, with a lot of blood fat sample and very thick blood can not be used.6. The operator can not touch the injection port and chromatography membrane at
detection port during test procedure.
7. The waster during the test procedure should be dispose as contamination according
to related regulations.
